So from the 6 ranked and 3 practice games I have played here are my initial reactions:

If you played dota on wc3 then you will find very little difference. Improvements are:

-The shop is only one person and all recipes are in that one shop
-The secret shops are more outlined and items tell you if you need secret shop or not.
-The heroes got new models and feel more solid.
-Improved graphics obviously.

What stayed the same:
-Most of the items
-The heroes are all the same, maybe some abilities changed but I'm not sure.

What DotA does wrong:
-It has picked up very few things, the biggest of all is a surrender button. It is really annoying having to play out 20 more minutes of a losing match.
-Nothing new:
Gameplay wise everything is generally the same(keep in mind last time I played DotA was 4 years ago) However with games such as HoN, and LoL picking up and surpassing the title of DotA clone by adding new gameplay mechanics. Think of it like this:

Megaman 1-8 are great games but fairly straightforward. By the 8th game people got very tired of it. So here comes megaman X which introduced dash and wall jumping. Suddenly they can get away with more games because the games feel like a completely different game.

DotA does not introduce any new gameplay mechanics which to me, holds it back. At the moment, to me, the difference between DotA and LoL is that:
DotA has a better community, nostalgia factor, and its new. Unless some new changes come it dethroning the beast that is LoL seems slim.

Of course I could be talking completely out of my ass, but these are just my first impressions of it and i look forward to playing more of it once finals are done.

Remember its still in beta too, there could be a lot to this game that they aren't showing us yet

1. I think valve is going to get dota 2 up to par with dota 1 before they start adding new heroes/mechanics
2. I prefer dota 1 gameplay already to lol, so dota 2 just being dota is good enough for me, the only thing I'm interested in is them having a surrender option,cleaning up the ui, and making it easier for beginners to get into.

Oh yeah on the Lol thing, I couldn't agree more. A surrender option would be nice, although they have already done quite a bit of cleaning up on the UI and the way shops work now as well as the suggested items really helps new players a lot

I never played Dota1, I just got introduced into the game style with LoL. Can one of you guys highlight the big differences between them?

Ok DotA1 was built on the wc3 engine so it was limited:
-The shops were scattered between like 7 different NPCs
-The secret shops weren't obvious and until someone told you about them, chances are you never saw them.
-DotA1 had friendly fire last hitting of creep and towers to deny gold.

HoN and LoL come out:
-They simplify the shop
-LoL gets rid of the friendly deny (with the exception of original gang plank) which supposedly makes games faster.
-LoL adds in talent specs, runes, summoner spells, and the bush, and more incentives to jungle
-Can't say much about HoN since I never got into it.

DotA2:
-Simplified shop
-updated graphics
-everything else from dota1 came over.

diffrences between dota and lol are this
-in dota you lose gold upon death
-every 2 minutes in dota runes spawn in the river they give buffs that range from invis,dd,regen,clones
-dota has champ mechanics that they wont put in lol, like perm pet heros and illusion heroes
-dota's spells don't scale with spells but do more damage without items so casters are powerful early-mid
-dota doesn't have free teleport back to base, instead you can buy scrolls that can use on any tower, they also have a courier to help you not leave your lane
-dota's towers are weaker allowing for easier ganks

Does DotA2 have leveling perks or unlocks that make higher level players significantly better than lower level players or is skill mostly based on actual character strategy?

It resets at every game. From the moment the game starts you have to play as close to perfectly as you can. The closer you get, the better you do, and that all shows up at the end. MOBA games are basically a war of attrition between two teams. If no characters die and towers don't fall, both teams have equal theoretical max income (killing all the neuts in the mobs and farming the jungles). That is why a character death or tower loss is a big deal because it puts the other team ahead in the net team-income race

*Edit*- I'm not really sure how relevant any of that is
